To get Clé des étages supérieurs you need reach Eye of the Jailer Tier 3/5 on The Maw by completing World Quests and Killing Elite/Rares.
When you reach Eye of the Jailer Tier 3/5 you gain the debuff Pourchassé.
When you are being hunted, an assassin will strike when you next enter combat.
This Assassin will drop the Clé des étages supérieurs (i got on my first kill). Note: the assassin drop another items, for example: & Témoignage ombre-lié.
After get Clé des étages supérieurs just complete the quest (auto pop).

Worth noting the daily quests you get from Ve'nari each grant a full Jailer tier.
